Enzyme engineering allows to explore sequence diversity in search for new properties. The scientific literature is populated with methods to create enzyme libraries for engineering purposes, however, choosing a suitable method for the creation of mutant libraries can be daunting, in particular for the novices. Here, we address both novices and experts: how can one enter the arena of enzyme library design and what guidelines can advanced users apply to select strategies best suited to their purpose? Section I is dedicated (...) to the novices and presents an overview of established and standard methods for library creation, as well as available commercial solutions. The expert will discover an up‐to‐date tool to freshen up their repertoire (Section I) and learn of the newest methods that are likely to become a mainstay (Section II). We focus primarily on in vitro methods, presenting the advantages of each method. Our ultimate aim is to offer a selection of methods/strategies that we believe to be most useful to the enzyme engineer, whether a first‐timer or a seasoned user. (shrink)

In an attempt to address the theoretical gap between linguistics and philosophy, a group of semanticists, calling itself the Generic Group, has worked to develop a common view of genericity. Their research has resulted in this book, which consists of a substantive introduction and eleven original articles on important aspects of the interpretation of generic expressions. The introduction provides a clear overview of the issues and synthesizes the major analytical approaches to them. In the past couple of decades, interest in the philosophy of language has, according to the introduction written by Zoltan Szabó for this volume, cooled somewhat. This is because pursuit of many of the philosophical issues that the study of language had set out to solve in the first place has developed into not so much the study of language, but, rather, of mind, and adjacent areas of philosophy. As Tyler Burge has put it, “Many philosophers felt that the philosophy (...) of language had done its job.” This volume attempts to illuminate the positive side of these developments for the philosophical study of language. Szabó says, “The scientific results are interesting on their own, their relevance to the philosophical questions remains clear”. This, then, puts philosophy of language on a new footing, where, “Freed from the responsibility of ultimately answering all philosophical questions, philosophers of language were free to confess an interest in language per se”. The stated purpose of the volume is to present the reader with a variety of representative papers which “exemplify the current reorientation of philosophy of language,” which Szabó characterizes as a joint linguistic and philosophical investigation into “the syntax and semantics of various natural language constructions.”. In (1991), Meinwald initiated a major change of direction in the study of Plato’s Parmenides and the Third Man Argument. On her conception of the Parmenides , Plato’s language systematically distinguishes two types or kinds of predication, namely, predications of the kind ‘x is F pros ta alla’ and ‘x is F pros heauto’. Intuitively speaking, the former is the common, everyday variety of predication, which holds when x is any object (perceptible object or Form) and F is a property (...) which x exemplifies or instantiates in the traditional sense. The latter is a special mode of predication which holds when x is a Form and F is a property which is, in some sense, part of the nature of that Form. Meinwald (1991, p. 75, footnote 18) traces the discovery of this distinction in Plato’s work to Frede (1967), who marks the distinction between pros allo and kath’ hauto predications by placing subscripts on the copula ‘is’. The abilities to attribute an action to its proper agent and to understand its meaning when it is produced by someone else are basic aspects of human social communication. Several psychiatric syndromes, such as schizophrenia, seem to lead to a dysfunction of the awareness of one’s own action as well as of recognition of actions performed by other. Such syndromes offer a framework for studying the determinants of agency, the ability to correctly attribute actions to their veridical source. Thirty normal (...) subjects and 30 schizophrenic patients with and without hallucinations and/or delusional experiences were required to execute simple finger and wrist movements, without direct visual control of their hand. The image of either their own hand or an alien hand executing the same or a different movement was presented on a TV-screen in real time. The task for the subjects was to discriminate whether the hand presented on the screen was their own or not. Hallucinating and deluded schizophrenic patients were more impaired in discriminating their own hand from the alien one than the non-hallucinating ones, and tended to misattribute the alien hand to themselves. Results are discussed according to a model of action control. A tentative description of the mechanisms leading to action consciousness is proposed. (shrink)

Several different evaluation issuesare perceived as important by people involved withinnovative projects intended to improve local food andnutrition systems; particularly the establishment oflocal food policy coalitions. Several such coalitionshave been formed in North America, Europe, andAustralia with the goal of improving community foodsecurity and promoting sustainable local food systems.Pioneer coalitions have served as models, yet therehas been little systematic evaluation of thesemodels. A qualitative study was conducted to identifyfactors that may hinder evaluation efforts. In grouptelephone interviews, we sought the views (...) ofacademics, project organizers, and funders, a total of24 key informants. Pressures to evaluate were assessed differently bythe three groups of key informants. Academics felt thefocus of evaluation should be on the effectiveness ofthe process used to discuss issues and formulatepolicies and plans. Project organizers and fundersperceived a need to assess project impact andoutcomes. A lack of suitable evaluation models andmethods was viewed as a formidable barrier. The use ofinappropriate methods and premature impact evaluationwere noted as potential threats to projectsustainability. External constraints and resourcelimitations were also said to inhibit evaluationefforts. It appears that several other factors may also beimpeding progress in conducting more (and more useful)evaluations including: (1) the apparent negativeconnotation of evaluation and the limited benefitsexpected from evaluation by stakeholders, (2) a lackof consensus about important evaluation questions,(3) insufficient evaluation expertise among projectorganizers, and (4) inadequate appreciation ofincreasing accountability pressures. Some w o u l d count such issues as h o w to e f f i c i e n t l y i n d e x databases as part of the philosophy of automated theorem p r o v i n g . (...) Others wonder about whether f o r m u l a s should be represented as strings or as trees or as lists, and call this part of the philosophy of automated theorem p r o v i n g . Yet others concern themselves w i t h what k i n d o f search should b e embodied i n a n y automated theorem prover, or to what degree any automated theorem prover should resemble Prolog. Still others debate whether natural deduction or semantic tableaux or resolution is " b e t t e r " , a n d c a l l t h i s a part of the p h i l o s o p h y of automated theorem p r o v i n g . Some people wonder whether automated theorem p r o v i n g should be " h u m a n oriented" or "machine o r i e n t e d " — sometimes arguing about whether the internal p r o o f methods should be " h u m a n - I i k e " or not, sometimes arguing about whether the generated proof should be output in a f o r m u n d e r s t a n d a b l e by p e o p l e , and sometimes a r g u i n g a b o u t the d e s i r a b i l i t y o f h u m a n intervention in the process of constructing a proof. Le déterminisme implique que le libre arbitre n’existe pas, que nous ne pouvons pas faire autrement ; réciproquement, avoir la possibilité de faire autrement implique que le déterminisme ne s’applique pas à l’instant, s’il existe, où on l’exerce. Cependant, la question de la responsabilité rend difficile d’accepter que les agents ne puissent pas faire autrement et motive fortement à rendre compatibles déterminisme et libre arbitre ou à soutenir, dans une veine « incompatibiliste », que le cerveau humain n’est pas soumis (...) à la loi du déterminisme. Le libertarianisme, c’est-à-dire l’idée que, chez l’agent humain, il existe des exceptions au déterminisme causal lors de l’activité cérébrale, n’est pas défendable à la lumière des découvertes récentes sur la cognition humaine. Selon le compatibilisme, un agent n’ayant pas la possibilité de faire autrement jouit encore de son Libre Arbitre. La théorie de Harry Frankfurt d’une volonté à plusieurs niveaux est étudiée de manière critique. On présente la preuve neuroscientifique d’un modèle de volonté en cascade, dans l’esprit de Frankfurt. On a souvent souligné que l’usage du concept de substance était problématique dans les Méditations, préférant l’exposé plus rigoureux des Principia. À l’encontre de cette interprétation, cet article affirme que le traitement analytique de la substance est non seulement conforme à l’ordre des raisons, mais que s’y développe une aventure révélatrice : si la réflexion sur l’ ego et la cogitatio tend à faire de celui-ci une quasi substance, la formule « ego autem substantia » accomplit dans la Troisième Méditation (...) une substantialisation qui n’est que transitoire. Car cette substantialisation, fondée sur la présomption de l’ ego se concevant comme author, est aussitôt contestée à la lumière de la relation que l’ ego entretient au Dieu infini. (shrink)

Employee attributions and emotional reactions to unethical behavior of top leaders in an organization recently involved in a highly publicized ethics scandal were examined. Participants (n = 76) from a large southern California government agency completed an ethical climate assessment. Secondary data analysis was performed on the written commentary to an open-ended question seeking employees' perceptions of the ethical climate. Employees attributed the organization's poor ethical leadership to a number of causes, including: lack of moral reasoning, breaches of trust, hypocrisy, (...) and poor ethical behavior role modeling. Emotional reactions to corruption included cynicism, optimism, pessimism, paranoia and fear, and were targeted at top leaders, organizational practices (i.e., the old boy network, nepotism, and cronyism) and ethics interventions. Implications for leadership training and other organizational ethics interventions are discussed. (shrink)

Leibniz n’a traité nulle part systématiquement de l’attention. Pourtant, en suivant le fil conducteur de l’attention comprise comme réflexion à soi et à ses propres actes, cet article montre qu’elle soulève différents problèmes liés à la constitution progressive du lexique de l’aperception. Le problème éthique d’un défaut d’attention amène Leibniz à reconnaître un mode non réfléchi de celle-ci, à en trouver le ressort naturel dans la constitution du corps percevant, et à la distinguer de l’aperception comprise comme réflexion à ce (...) qui est en soi : non pas réflexion morale sur ses propres actes, mais saisie des idées innées et de leur liaison, qui fondent la personnalité morale. Il faut alors refuser l’interprétation récente qui propose d’étendre l’aperception aux âmes animales sous prétexte qu’elles auraient de la sensation : dans la langue philosophique de Leibniz, le chien s’aperçoit de sa douleur et peut y prêter attention ; il n’en a pas d’aperception. Henri Bergson, Vladimir Jankélévitch, Emmanuel Levinas ont en commun d'avoir marqué la philosophie européenne du XXe siècle. En plus de leurs oeuvres respectives, ils ont tous les trois entretenu des liens manifestes à la fois dans leurs textes et dans leurs biographies. De ce trio, on ne peut pas dire que Bergson, philosophe pourtant déjà installé dans le paysage intellectuel français, prenne la place privilégiée du maître. Ni Jankélévitch, ni Levinas n'ont été, à proprement parler, ses élèves, cependant une relation (...) étroite s'est instaurée entre eux, marquée non pas par une posture de déférence, mais par des préoccupations conceptuelles partagées ainsi qu'un travail engagé sur des questions communes. Les pensées de Jankélévitch et Levinas sont liées par un même franc retrait vis-à-vis de la majorité des courants philosophiques dominants à leur époque, notamment la vogue existentialiste. N'adhérant pas à cette tendance, Jankélévitch et Levinas se tournent vers Bergson pour saisir l'inactualité des questions posées par sa pensée ; plutôt que la restitution de ses thèses, leurs interactions reposent sur un véritable dialogue dans lequel chacun apporte une variation nouvelle. Ces trois philosophes seront finalement réunis par le partage de questions fondamentales, qui résistent à la compréhension et qui s'ouvrent au souci de l'expérience proprement humaine : métaphysique, morale, temps. Bergson, Jankélévitch et Levinas, trois philosophes français marqués par une même inquiétude face à la dégradation de l'humain ; trois moments qui forment une ligne de pensée discrète mais irradiant la philosophie contemporaine, révélée après-coup par les suites qu'elle a autorisées. Dans son compte-rendu de mon livre, Les Animaux Pensent-ils?, Machery objecte que l'évolution n'étant ni hiérarchique ni linéaire, il n'et pas justifié de proposer une analyse hiérarchique des représentations. Je réponds à cette objection, en montrant qu'on peut en effet distinguer des types de représentation par leurs propriétés sémantiques et computationnelles. On peut reconnaître le caractère anagénétique du développement de la cognition sans pour autant légitimer une conception hiérarchique et continuiste de l'évolution des espèces.

Cet article expose l’intérêt qu’il peut y avoir pour la recherche à éditer les notes de lecture de Leibniz dans les Sämtlichen Schriften und Briefen. Il examine à titre d’exemple un mémoire sur la politique sociale et économique que Leibniz a soumis à la cour de Prusse en 1701, deux notes de lecture sur des propositions semblables faites par d’autres ainsi que deux extraits des œuvres de Charles Davenant. Leur comparaison permet entre autres de montrer d’où Leibniz tenait ses informations (...) et comment il se rapportait à son environnement intellectuel. Dans la mesure où Leibniz formule à l’occasion de la lecture de Davenant des considérations fondamentales sur l’économie qui n’ont pas d’équivalent ailleurs dans le corpus, cet article s’emploie en même temps à saisir le rapport de Leibniz aux économistes anglais et à situer plus précisément ses conceptions dans l’histoire des idées. La plupart des philosophes de l’Antiquité pensent que les rêves ont des capacités prophétiques. Les seuls philosophes à s’écarter de cette tradition sont les épicuriens. Ils démystifient le phénomène du rêve en l’expliquant dans les termes de leur théorie physique matérialiste. Ils soulignent le lien entre le contenu du rêve et les activités diurnes, entre ce même contenu et la qualité d’âme du rêveur. Parce que l’identité de chacun persiste lorsqu’il rêve, les épicuriens attribuent à l’activité onirique une signification psychologique (...) et morale considérable : quoique non véridique, le rêve n’en reste pas moins révélateur. Cet article examine le traitement donné par Descartes et certains de ses successeurs d’une question classique, quoique peu étudiée, celle de savoir si l’on peut penser plusieurs choses à la fois. Le thème d’une saisie simultanée est central dans la théorie cartésienne de la connaissance, en particulier dans les Regulæ, où il s’appuie sur le recours à une attention divisée. Restreignant ce pouvoir à la seule imagination, Clauberg voit dans le corps vivant le paradigme de la simultanéité. Spinoza offre quant (...) à lui un point de vue synthétique : la simultanéité dans le corps et l’esprit sont des expressions de la puissance individuelle et de son progrès. L’attention n’est plus comme chez Descartes une condition de la simultanéité intellectuelle, mais un obstacle qu’il s’agit de surmonter pour comprendre plus et mieux. (shrink)

Cette présentation au volume Lectures de Martial Gueroult a pour objet de redonner le cadre général dans lequel Gueroult a défendu sa méthode en histoire de la philosophie. Plus que l’opposition entre genèse et système, ce sont plutôt les manières différentes de comprendre ce qu’est un héritage philosophique qui ont structuré le débat. Gueroult a cherché à restituer ce que les œuvres lèguent d’elles-mêmes en héritage, par une lecture interne ou verticale, qui ne les juge ni par leur inscription horizontale (...) dans une série d’œuvres antécédentes, ni par les héritages revendiqués par la postérité. En considérant qu’un texte philosophique n’est pas le témoin d’une intuition mais le porteur d’une méthode propre, ce n’est pas seulement à Ferdinand Alquié qu’il s’est opposé, mais à toute une tradition française dont il identifie la pointe chez Henri Bergson. Si Leibniz a souvent présenté une analogie entre la substance et l’ ego, la détermination de la substantialité à partir de l’ ego ne relève cependant ni d’une analogie, ni d’un transfert de substantialité ni d’une « déduction égologique ». Il s’agit plutôt d’une hypotypose, ancrée dans l’expérience du sujet en première personne, qui est toujours l’expérience d’un corps associé. Sans cette expérience réflexive, le concept même de substance, qu’il soit identifié à la subjectité ou à la perséité, n’aurait aucun (...) sens. Par cette expérience réflexive, c’est toujours l’expérience d’un corps substantié et d’une infinité de substantiés qui est en jeu. ObjectivesPrevious studies found a general increase in prejudice against Chinese people during the first months of the pandemic. The present study aims to consider inter-individual heterogeneity in stability and change regarding prejudice involving Chinese people during the pandemic. The first objective is to identify and describe different trajectories of prejudice over a seven-month period during the pandemic. The second and third objectives are to test the association between trajectory group membership and antecedent variables such as: socio-demographic factors (i.e., age, gender, (...) political affiliation) and two psychological mechanisms, namely economic threat and global citizenship identification.MethodsA representative Canadian sample (N = 3,617) according to age, gender and province of residence, was recruited for a 10-wave survey starting from April 2020 to December 2020. First, a group-based modeling approach was used to identify trajectories of prejudice. Second, a multinomial logistic regression model was used to test associations between membership in trajectories and antecedents.ResultsFour trajectories were identified. The first three trajectories have a low (71.4% of the sample), high (18.5%) or very high (5.3%) level of prejudiceagainstChinese people which is relatively stable over time. The fourth trajectory (4.9%) reports low levels of prejudicein favorof Chinese people which become more positive throughout 2020. Regarding socio-demographic factors: gender is not associated with trajectory group membership, younger people are more likely to follow the trajectory in favor of Chinese people and conservatives are more likely to follow the highest trajectories against Chinese people. Regarding some psychological mechanisms: personal but not collective economic threat is associated with the trajectory in favor of Chinese people. Finally, the highest levels of prejudice are found when the strategy of identification is more local rather than global.ConclusionThe present study shows that Canadians differ in terms of both their level and change in prejudice against Chinese people throughout the pandemic with some socio-demographic groups being more likely than others to be associated with prejudice. The results also suggest that a promising way to tackle the major social issue of prejudice is to highlight a vision of the world where individuals are all “global citizens” facing the same challenge. Fuzzy logics are systems of logic with infinitely many truth values. Such logics have been claimed to have an extremely wide range of applications in linguistics, computer technology, psychology, etc. In this note, we canvass the known results concerning infinitely many valued logics; make some suggestions for alterations of the known systems in order to accommodate what modern devotees of fuzzy logic claim to desire; and we prove some theorems to the effect that there can be no fuzzy logic which (...) will do what its advocates want. Finally, we suggest ways to accommodate these desires in finitely many valued logics. (shrink)

We investigate the notion of relevance as it pertains to ‘commonsense’, subjunctive conditionals. Relevance is taken here as a relation between a property (such as having a broken wing) and a conditional (such as birds typically fly). Specifically, we explore a notion of ‘causative’ relevance, distinct from ‘evidential’ relevance found, for example, in probabilistic approaches. A series of postulates characterising a minimal, parsimonious concept of relevance is developed. Along the way we argue that no purely logical account of relevance (even (...) at the metalevel) is possible. Finally, and with minimal restrictions, an explicit definition that agrees with the postulates is given. (shrink)

T. K. Menas [4, pp. 225-234] introduced a combinatorial property χ (μ) of a measure μ on a supercompact cardinal κ and proved that measures with this property also have the partition property. We prove here that Menas' property is not equivalent to the partition property. GEORGE adopts from Dionysius.4 It is a word that has different but related meanings in the moral, intellectual, and spiritual orders. Our intention is to elucidate the thomistic doctrine of ' leadingby -the-hand ' in the context of intellectual education, or ' mind forming,' discussing its nature, its necessity, and its place in the global picture of human knowing as outlined by Aquinas. We will begin by delineating in general what manuductio is by comparing two key passages, the first of which is to be found in the Summa Theologiae: The teacher leads the students from what is already known to knowledge of things unknown in two ways. First, by putting before him certain aids or instruments which his intellect uses in order to acquire science ; for example, when he presents him with some less universal proposition which nevertheless the student is able to judge from things already known; or when he proposes to him some sensible examples, or similitudes, or opposites, or some other things of this sort, from which the intellect of the learner is ' led by the hand ' (manuducitur) to the knowledge of a truth previously unknown to him. The other way [the teacher leads the student] is when he strengthens the intellect of the learner... inasmuch as he proposes the order of principles to conclusions to the student, who perhaps by himself would not have so much ability to put things together (virtutem collativam) that from the principles he could deduce the conclusions. And therefore it is said in Posterior Analytics. Bk. I, that 'demonstration is a syllogism making one know '. And through this mode the one who demonstrates makes the listener know.5 4 Cf. Haricîlere göre ise, ister büyük, isterse küçük olsun, her türlü günah sahibini iman dairesinden çıkarır ve küfür dairesine dahil eder. Molla Halil’e göre, ahiret hayatı ruh ve cesetle birlikte olacaktır. Bu üm-metin günahkârlarına peygamberler, şehitler ve salih kimseler şefaat edecek-tir. Mu‘tezile‘ye göre şefaat sadece iyilerin derecesinin yükselmesi içindir. Günahkârlar için şefaat olmayacaktır. Günahkâr müminler Cehenneme girseler de ebedi kalmayacaklar. Kâfirlerin azapları ise devamlı olacak ve bunlar ebedi olarak Cehennemde kalacaklardır. Biz bu makalede önce sevap ve İkâb kelimelerinin lügat ve ıstılah anlamları üzerinde duracak, sonra bu bağlamda ayet ve hadislerde verilen bilgileri ve bunların açıklamalarını ele alacağız. Daha sonra Molla Halil’in bu konulara dair görüşlerini sunmaya çalışacağız. Onun sevap ve İkâb ile ilgili tanımlarını, bunların sonuçlarına dair yaşanacakları ve varılacak yerleri ele alan yaklaşımlarını sunmaya çalışacağız. Burada farklı görüşlere sahip olmaları nedeniyle çoğunlukla Mu‘tezile’nin ve az da olsa Haricilerin görüşlerine de mukayese oluşturması için değineceğiz.
